Peptide research and FDA decisions, in plain words. What happened, what it means, and where the evidence stops.

## The shortage ended. Local gaps could still happen.

Event · 21 February 2025

FDA chronology updated · 1 April 2026

FDA said national supplies of semaglutide injections could meet demand. That changed the rules for compounded copies, too. A supply decision and a drug approval are still different things.

## What the end of the shortage changed

Event · 21 February 2025

FDA chronology updated · 1 April 2026

### What happened

On 21 February 2025, FDA [declared the national shortage of semaglutide injections over](https://www.fda.gov/drugs/drug-alerts-and-statements/fda-clarifies-policies-compounders-national-glp-1-supply-begins-stabilize). Manufacturing could meet U.S. demand, the agency said. That didn’t mean every local shelf would stay stocked.

Tirzepatide had its own timeline: an initial decision on 2 October 2024, then a new decision on 19 December after another review. The two medicines didn’t leave the shortage list together.

### What it means

The shortages had allowed certain compounded copies: medicines prepared by a pharmacy or another authorized facility, rather than an approved finished product. Once national supplies recovered, those shortage-based exceptions ended after temporary grace periods.

For semaglutide, the original pharmacy deadline was 22 April 2025, or the court’s ruling if later. FDA confirmed that period had ended after the 24 April ruling. Larger outsourcing facilities had until 22 May. The [regulatory page](/regulatory) puts those dates in order.

Neither semaglutide nor tirzepatide has FDA approval in compounded form. FDA has [reported medication errors with compounded semaglutide](https://www.fda.gov/drugs/drug-alerts-and-statements/fdas-concerns-unapproved-glp-1-drugs-used-weight-loss), including some requiring hospital care. A licensed U.S. clinician can discuss approved treatments and an individual’s needs.

## Research

### Retatrutide results came from two different groups

Event · 26 June 2023

ADA announcement · 26 June 2023

#### What happened

On 26 June 2023, the American Diabetes Association [announced two retatrutide trials](https://diabetes.org/sites/default/files/2023-10/ada_sci_sessions_2023_news_release_retatrutide_0.pdf). These were phase 2 studies: mid-stage human testing. One studied adults without type 2 diabetes; the other studied adults with it.

#### What it means

The [obesity trial](https://pubmed.ncbi.nlm.nih.gov/37366315/) found weight loss, alongside stomach side effects and a faster heartbeat. Retatrutide is an unapproved investigational drug. [FDA says it cannot be compounded](https://www.fda.gov/drugs/drug-alerts-and-statements/fdas-concerns-unapproved-glp-1-drugs-used-weight-loss) under federal law; a trial result doesn’t change that.

### Less liver fat in a study of adults with HIV

Event · 15 October 2019

NIH announcement · 15 October 2019

#### What happened

On 15 October 2019, NIH [reported a year-long tesamorelin trial](https://www.nih.gov/news-events/news-releases/drug-reverses-signs-liver-disease-people-living-hiv) in adults with HIV and fatty liver disease. Liver fat fell more with tesamorelin than with placebo, a comparison treatment without the medicine.

#### What it means

That finding doesn’t make tesamorelin a general weight-loss treatment. Its [prescribing information](https://dailymed.nlm.nih.gov/dailymed/drugInfo.cfm?setid=839334d3-8c1d-4c26-9036-2ab524a6ea75) warns about blood sugar and fluid retention. The [full report](/tesamorelin) explains what was studied and what is approved.
